Vitronectin receptor (alpha(V)beta(3)) antagonism has been implicated as a mechanism for the treatment of restenosis following balloon angioplasty. In this work we present results from screening of a focused combinatorial library based on a biphenyl moiety. Our SAR studies led to the identification of compounds with subnanomolar activity, selectivity towards the related GPIIbIIIa receptor and functional activity on human smooth muscle cell migration.  相似文献

In an effort to identify widely active positive regulatory elements, we have examined the action of the cytomegalovirus enhancer-promoter in transgenic mice. These elements activated expression in 24 of 28 tissues tested. The greatest expression was observed in the heart, kidney, brain, and testis. Maximum expression further localized to specific cells within the heart and kidney.  相似文献

The Escherichia coli secA gene, whose translation is responsive to the proficiency of protein export within the cell, is the second gene in a three-gene operon and is flanked by gene X and mutT. By using gene fusion and oligonucleotide-directed mutagenesis techniques, we have localized this translationally regulated site to a region at the end of gene X and the beginning of secA. This region has been shown to bind SecA protein in vitro. These studies open the way for a direct investigation of the mechanism of secA regulation and its coupling to the protein secretion capability of the cell.  相似文献

'15N signatures of fossil peat were used to interpret past ecosystem processes on tectonically active subantarctic Macquarie Island. By comparing past vegetation reconstructed from the fossil record with present-day vegetation analogues, our evidence strongly suggests that changes in the '15N signatures of fossil peat at this location reflect mainly past changes in the proportion of plant nitrogen derived from animal sources. Associated with uplift above sea level over the past 8,500 years, fossil records in two peat deposits on the island chronicle a change from coastal vegetation with fur and elephant seal disturbance to the existing inland herbfield. Coupled with this change are synchronous changes in the '15N signatures of peat layers. At two sites 15N-enriched peat '15N signatures of up to +17‰ were associated with a high abundance of pollen of the nitrophile Callitriche antarctica (Callitrichaceae). At one site fossil seal hair was also associated with enriched peat '15N. Less 15N enriched '15N signatures (e.g. -1.9‰ to +3.9‰) were measured in peat layers which lacked animal associated C. antarctica and Acaena spp. Interpretation of a third peat profile indicates continual occupation of a ridge site by burrowing petrels for most of the Holocene. We suggest that 15N signatures of fossil peat remained relatively stable with time once deposited, providing a significant new tool for interpreting the palaeoecology.  相似文献

An isometric virus was isolated from Helianthus annuus L. plants showing a yellow leaf spot mosaic on affected leaves. Infected plants were found in different ecological regions of Ukraine. A procedure of virus purification is described. The diametres of the virus particles were nonuniform and ranged from 50 to 120 nm. The sedimentation coefficient of the virus was 518–540 S and the floating density in the CsCl gradient was 1.22 g/cm3. The MW of proteins separated by electrophoresis amounted to 78±0.9, 58±0.8, 52±0.2, and 27±0.8 kDa, respectively. The virus was assigned to the tospoviruses for which sunflower is a new previously undescribed natural host plant.  相似文献

An enzymatic activity associated with intracellular membrane fractions of Merwin plasma cell tumor II, baby hamster kidney, and chicken embryo fibroblast cells and bovine kidney has been characterized which covalently links fatty acids onto the G protein of vesicular stomatitis virus. Exogenous G protein extracted from native vesicular stomatitis virus particles can be acylated in vitro only after it has been previously deacylated. The fatty acids transferred in vitro are sensitive to treatment with hydroxylamine, indicating an ester linkage. Cell-free acyl transfer was also observed with endogenous G protein present in membrane fractions prepared from vesicular stomatitis virus-infected cells. In this case, the fatty acids become linked to a G protein species (G1) which is not terminally glycosylated and therefore has not entered the trans-Golgi compartment. The same G protein species also becomes acylated in infected cells during short pulses with radioactive palmitic acid. Acylation of the G protein in vitro with free palmitic or myristic acid is energy-dependent, and the addition of ATP is specifically required. Other nucleoside triphosphates cannot substitute for ATP in the activation of free acyl chains. Alternatively, activated fatty acids linked in a high energy thioester bond to coenzyme A, e.g. [14C] palmitoyl-CoA, are suitable lipid donors in the in vitro acylation reactions. Palmitic acid transfer onto G protein shows the typical characteristics of an enzyme-catalyzed reaction.  相似文献
